Eine besondere Rolle spielt in diesem Spektrum von Aufgaben das Problem der stabilen Schwimmfähigkeit eines Körpers im Wasser. Die klassische Stabilitätstheorie ist auf der Basis von hydrostatischen Reaktion gegründet. D.h. es werden Verdrängung, Metahöhe und Metaradius des Schiffes bestimmt, und auf dieser Basis wird eine Stabilitätsbedingung formuliert. In diesem Werk wird eine Methode entwickelt, die eine bestimmte Verallgemeinerung von klassischer Technik darstellt. Die Methode gilt sowohl für schwimmende (auf der Wasseroberfläche) als auch für schwebende (unter der Wasseroberfläche) Körper. Die Verallgemeinerung der klassischen Methode wurde in zwei Richtungen vorgenommen. Die erste Richtung: Der Körper wird als mehrfach zusammenhängendes Gebilde dargestellt. Es wird eine multikomponente Darstellung vom Körper bezüglich Dichte eingeführt, d.h. der schwimmende Körper wird in seine Bestandteile (z.B. Stahlkonstruktion, Ladung, Luftraum, Ballastwasser) zerlegt. Auf dieser Basis wird eine multiparametrische Darstellung vom Körper-Wasser-System in Matrizenform eingeführt. Die Grundlage dieser Darstellung bilden Vermessungs-, Massen- und Verdrängungsmatrix. Diese 6×6 Matrizen enthalten geometrische, mechanische und hydrostatische Momente vom Körper. Sie beschreiben bestimmte Integralwerte (Funktionale) vom Körper-Wasser- System: Skalar-, Vektor- und Tensormoment. Grundlage für diese Integralwerte bilden entsprechende Volumen-, Dichten- und Verdrängungsverteilungsfunktionen. Die Schwimmmatrix wird als Kombination von o.g. Matrizen konstruiert. Sie beschreibt das Maß von Schwimmfähigkeit und -Stabilität des Körpers in Form von Volumenverhältnissen zwischen Körperbestandteilen. Die Schwimmmatrix lässt ihre Vektor- und Tensorkomponenten zu anderen Koordinatensystemen umformen. Damit entsteht die Möglichkeit, die Schwimmfähigkeit und -Stabilität vom Körper in Abhängigkeit von allen denkbaren Variationen von geometrischen und mechanischen Parametern zu untersuchen. Die zweite Richtung: Die Anfangsstabilität eines schwimmenden (bzw schwebenden) Körpers wird als Stabilität des Gleichgewichtszustandes definiert. Es wird das Gleichgewichtszustand des Körpers im Wasser analysiert. Das hydrostatische Kraftsystem eines Körpers (H-Kraftsystem) wurde eingeführt, das eine Kombination von Auftriebskräften und zusammenpressenden Belastungen darstellt. Das H-Kraftsystem bildet ein vollständiges System von Vektoren, das den Gleichgewichtszustand des Körpers beschreiben lässt. Das H-Kraftsystem wird mit einer Matrix (H - Matrix) erfasst, die drei Kräfte und drei Drehmomente beinhaltet. Die Komponenten der H-Matrix (H-Funktionale) hängen von mehreren Umständen ab: der geometrischen Form des Körpers, der Tauchtiefe, der Orientierung des Körpers bezüglich der Wasseroberfläche (bzw Schwerkraftrichtung). Auf dieser Grundlage wurden die aufrichtenden Drehmomente bezüglich Krängung und Trimm konstruiert. Sie bestimmen das Maß von Stabilität des Körpers bezüglich der äußeren Einwirkungen. Light refractive tomography (LRT), which utilizes laser light to detect ultrasound pressure in fluids, has long been underestimated as a simplification of another optical technique, light diffraction tomography (LDT). After a thorough review of published research, it appears that LRT is so far the sole technique, which is noninvasive, omnidirectional and provides time-resolved results scaled in Pa. In the process of developing LRT into a reliable and useful ultrasound measurement technique, achievements have been made in four different aspects: - Accuracy: With the aid of numerical simulations, potential systematic errors are quantitatively analyzed. Weiterhin wird erstmalig von der plasmonischen Kopplung von Nanopartikeln in einer dreidimensionalen optischen Falle berichtet und die Auswirkung der Kopplung auf die Stabilität der Falle untersucht. So genannte optische Pinzetten finden seit Jahrzehnten unter Anderem in der Laserkühlung, der Erzeugung von Bose-Einstein-Kondensaten und der Spektroskopie breite Anwendung. Mit ihnen können Objekte kontaktlos über lange Zeit immobilisiert werden. Daher sind sie auch für die Raman-Spektroskopie interessant, in der kleine Streuquerschnitte von Molekülen lange Integrationszeiten bedingen. Eine bekannte Methode, diese Aufnahmezeiten durch Nutzung der lokalen Feldüberhöhung in plasmonisch aktiven Systemen zu verkürzen (SERS), konnte aber bislang nicht in optischen Pinzetten umgesetzt werden. Im ersten Teil dieser Arbeit wird erstmals plasmonisches Koppeln optisch dreidimensional gefangener Silber-Nanopartikel beobachtet. Die Kopplung verstärkt sich mit der Zeit, bis Teilchen aus der Falle freigesetzt werden. Dieser Zusammenbruch der Falle wird mit Modellrechnungen untersucht. Dabei werden je in Abhängigkeit des Partikelabstands die Tiefe des Fallenpotentials und die Aufheizung des umgebenden Wassers durch plasmonisches Heizen untersucht. Thermophoretische Kräfte durch eine starke Temperaturzunahme um die Partikel herum können als Ursache für den Zusammenbruch der optischen Falle identifiziert werden. Bei genauer Kontrolle des Partikelabstands ist plasmonische Kopplung als Grundvoraussetzung für SERS damit in optischen Fallen prinzipiell möglich. Im zweiten Teil dieser Arbeit wird die Beobachtung der Dynamik optisch gefangener Gold-Nanopartikel genutzt, um akustische Spektroskopie auf der Mikroskala zu betreiben. Makroskopische Schallquellen können durch eine Veränderung der ansonsten durch Brownsche Bewegung dominierten Dynamik einfach identifiziert werden. Als mikroskopische Schallquellen kommen plasmonisch geheizte Aggregate von Gold-Nanopartikeln zum Einsatz. Sie verändern die Dynamik der beobachteten gefangenen Partikel im Zeitraum nicht signifikant. Erst Analysen im Frequenzraum können mikroskopische Schallquellen eindeutig nachweisen. Dabei kann die Schallquelle nicht nur spektroskopiert werden, auch ihre Richtung lässt sich bestimmen. Die messbare Schallleistung liegt in den durchgeführten Versuchen bei LW = -58 dB. Die dem detektierenden Partikel dabei zugeführte Energie liegt mit Ekin ~ 90 µeV bei nur etwa 0, 1% seiner thermischen Energie. Dank dieser neuartigen Technik können bislang nicht zugängliche Vorgänge in lebenden oder künstlichen Mikro- oder Nanosystemen mit einer unerreicht hohen Empfindlichkeit akustisch spektroskopiert werden. ····· 10361107430

However, recent investigations have brought this pathology into contention with significant evidence implicating subchondral bone in the disease`s pathophysiology. Early-stage primary osteoarthritic bone samples were harvested from both presenting medial and and non-presenting lateral compartments of diseased human tibia and were used to investigate the role of subchondral bone in the pathogenesis of primary human osteoarthritis. Significant differences in intrinsic tissue stiffness and mineralisation were observed in all samples presenting with clinically-defined, early-stage OA. Interstingly, non-presenting lateral samples also exhibited significant differences in stiffness and mineralisation. Hence, the presence of an early, non-presenting stage of OA initiation was observed in the bone tissue of these samples prior to overlying cartilaginous tissue damage. In conclusion, the classic `wear` and `tear` definition of this disease is no longer applicable to human knee OA. ····· 10361101924
