Economically importance genera Trichoderma is used more frequently in crop production and plant agriculture, for high yield and in disease management as biological control agents. In present study genetic characterization (rRNA gene amplification) of twenty four indigenous isolates of Trichoderma was carried out through these standard protocols. Biochemical analysis regarding different enzymes was also performed to identify the variability of isolates on the basis of their enzyme production potential. Nine hundred seventy Aspergilli strains were isolated from cellulosic wastes and twenty four strains were screened as cellulolytic and identified as Aspergillus tubengensis, A. niger, A. ficuum, A. awamori str. 1&2, A. foetidus, A. japonicus str.1&2, A. aculeatus str.1&2, A. flavus str.1,2,3 and 4, A. clavatoflavus, A. parasiticus, A. oryzae, A. spersus, A. fumigatus, A. cervinus, A. ochraceus, A. crustosus, A. terreus and A. cremeus. Among them A. oryzae showed the highest reducing sugar level and cellulase activity. The optimum pH, temperature and Km value of crude enzymes were 5.5, 50°C and 0.27%, respectively. The specific activity of purified extra and intracellular enzyme was 64.17 and 43.9 mg/ml although the increase of purification fold was about 53 and 41, respectively. The molecular weight was estimated to be 41,500 and 38,600 by gel filtration and 41,000 and 38,000 by SDS-PAGE. The enzymes gave single band and showed absorption spectra about 260-290 and 240-270 nm and Km value was 0.67% and 0.83%. The activities of the enzymes were increased in presence of Ca2+ , Mn2+ and Mg2+ but completely inhibited by 0.5M EDTA and significantly inhibited by Cu2+, Hg2+ and Fe3+. ····· 1036187395

The naphthalene dioxygenase (NDO) from Pseudomonas sp. Strain NCIB 9816-4 is a multicomponent enzyme system that carries out the initial step in the degradation of naphthalene. This enzyme has a broad substrate range and catalyzes several types of reactions including cis-hydroxylation, monooxygenation, and desaturation. In this study 1-phenylpyrazole and 1-phenylpyrrole were tested for biotransformation by NDO. HPLC analysis showed two metabolites from each substrate were produced by NDO. LC/MS analysis indicated the molecular weight of metabolites M1 and M2 of 1-phenylpyrazole were increased by 16 and 34 respectively which could be assumed as monooxygenation and dioxygenation reaction. On the other hand, HPLC and LC/MS analyses of metabolites M1 and M2 from 1-phenylpyrrole showed increased molecular weight of 16 and 32, as compared to the parent compounds, proposing addition of one and two atoms of oxygen to substrate, respectively. Since the early 1980s, methyl tert-butyl ether (MTBE) has been the most common octane booster and fuel oxygenate used in gasoline in the United States. The Clean Air Act of 1990 mandated the use of so-called reformulated gasoline by the late 1990s, widespread surface and groundwater MTBE contamination caused by gasoline spills and leakage became a crisis across the US. Two different microorganisms, Mycobacterium vaccae JOB5 and Graphium sp., were studied to evaluate their ability to aerobically cometabolize MTBE and its metabolites after growth on propane and iso-pentane. Both cultures were capable of cometabolizing MTBE and the metabolites, tert-butyl formate (TBF) and tert-butyl alcohol (TBA). MTBE, TBF, and TBA did not support growth of either microbe. Higher degradation rates were obtained in the bacterial system when the cultures were grown on iso-pentane. Nonlinear least squares regression and direct linear plot methods were used to estimate kinetic coefficients and provided comparable results. The enzymes involved in cometabolism of MTBE and its metabolites exhibited similar kinetics and substrate inhibition. Experimental data was modeled with Monod kinetics and STELLA®. ····· 1036186793

Giardia lamblia and Cryptosporidium parvum are parasitic protozoa that infect humans as well as domestic and wild animals all over the world. These parasites are implicated in many water borne disease outbreaks in different parts of the world. The present study was conducted to assess the prevalence of these two parasites among children below 14 years old that drink water from different sources. Single stool specimens were collected from a total of 384 children from the three villages. For identification of Cryptosporidium parvum the modified Ziehl-Neelsen staining method was used. Giardia lamblia was detected using direct microscopy based on wet mount and formalin-ether concentration techniques. Out of the 384 children examined, 102 (26.6%) and 31 (8.1%) were found positive for Giardia lamblia and Cryptosporidium parvum infection, respectively. Overall co-infection with intestinal parasites was detected in 4.4% of the study participants, in which Giardia lamblia and Cryptosporidium parvum comprised the highest proportion. ····· 1036186574

Peanuts are commonly infected with yellow molds or Aspergillus flavus during pre- and post-harvest stages, storage and transportation resulting in aflatoxin contamination. Consumption of aflatoxin contaminated peanuts causes health hazards like liver cancer, stunted growth and mental retardation, immune system suppression, allergies, and death in both humans and livestock. Recommended agronomic practices, good pre and post-harvest practices, chemical and biological control have been emphasized to overcome the above hazards. Adopting resistant varieties against Aspergillus flavus and aflatoxin contamination is considered as a cost effective strategy. Evaluation of different peanut varieties to A. flavus infection, determination of predominant Aspergillus species and levels of aflatoxin contamination in peanut varieties were the objectives of this study. Results showed that none of the varieties was immune to A. flavus infection. Differences were observed in the level of mycelial growth under different treatments. The Aspergillus species isolated from peanut kernels were A. niger, A. flavus, A. nidulans and A. fumigatus. The aflatoxin contamination levels ranged from 6 to 27 ppb. ····· 1036186558
