An investigation was carried out to study As, Cu, Fe, Mn, Pb and Zn contents and the hazards of these trace metal contamination in pond, lake and river waters of Dhaka metropolitan city. The average concentration of As, Cu, Fe, Mn, Pb and Zn were varied from 0.002 to 0.006, 0.030 to 0.413, 0.227 to 0.690, 0.173 to 0.900, 0.113 to 0.390 and 0.01 to 0.23 ppm, respectively. According to drinking water (potable water), public water, irrigation water and for aquaculture standard As, Cu, and Zn toxicity were not detected. The retina is a highly ordered laminar structure, comprising three compact layers of neurons separated by two synaptic layers, which has proven a valuable model to study gap junctions and cell specific expression patterns of connexins in neuronal tissues. Visual processing in the retina information is partly accomplished by laterally orientated horizontal cells. Two connexins, Cx52.6 and Cx55.5 has been shown to be expressed in zebrafish horizontal cells. Sequence analysis of zfCx55.5 and zfCx52.6 has revealed only limited homology of these connexins to other connexins from fish and higher vertebrates. ZfCx52.6 shows around ~57% amino-acid homology with zfCx55.5, where as the zfCx55.5 has been shown to share about 50% homology with the mouse Cx57. Both of these connexins have been found to show highly restricted expression in the horizontal cells of zebrafish retina. The molecular mechanism responsible for their cell specific expression are unveiled in this dissertation. Moreover, connexins perform various functions for which gap-junction communication seems to be dispensable. Plants are living factories which synthesize a large number of compounds as secondary metabolites, which are of immense economic value.The genus Sida belonging to the family Malvaceae is a group of angiosperm plants so far used for extraction of ephedrine, which is used in the treatments of rheumatism, heart diseases, troubles in urinary bladder and urethra and malaria. In vitro production of ephedrine from Sida calluses and methods to enhance the ephedrine content by inducing polyploidy was attempted in the present study. Callus was induced from Sida rhombifolia and Sida acuta using leaf explants in half MS medium supplemented with lmgl -1 NAA and 1mgl -1 2,4-D. For ephedrine production the calluses were grown in full MS medium supplemented with 2mgl -1 kinetin, 1mgl -1 NAA and 100mgl -1 L-phenylalanine.The ephedrine content in calluses of both diploid and colchiploid plants was analyzed using HPLC method. Ephedrine yields of colchiploids were 140 times higher than diploid control callus in Sida rhombifolia, while it was only 18 times higher in Sida acuta. In this study,prediction of physiochemical properties of catalytic sites residues using a suitable Artificial Neural Networking (ANN) Backpropagation algorithm coupled with a set of structural proteins with the properties of their amino acid residues. The method has been applied to a set of 100 structural proteins from the Protein Data Bank (PDB.Using Ligplot program for searching of active site residues the identified amino acid residues were classified in 15 different categories based on their physiochemical properties. After classification of active and non active site amino acids, their properties were converted into machine language. Furthermore, we created Neural Network Using Matlab software and generated algorithm for training and testing of data. Thereafter, analysis of results showed that 95% of active site s physiochemical properties were correctly predicted. It is hoped that this work would help in determining the surface topographic properties for ligand binding sites residues in protein. Hepatocyte nuclear factor 4 (HNF4 ) is a liver enriched transcription factor that belongs to the nuclear receptor family, and controls the expression of genes involved in various different functions, such as nutrient transport, metabolism, growth and differentiation. Recently, fatty acids and/or fatty acyl CoAs as well as some xenobiotics (e.g. hypolipidemic drugs) have been suggested as HNF4 ligands. LFABP was expressed and purified as a positive control, and used successfully to validate the DAUDA assay. Rat HNF4 LBD was successfully expressed and purified, therefore further work for ligand identification was performed using the rat HNF4 . Suitability of the DAUDA fluorescence displacement assay for screening of ligands for rat HNF4 was confirmed by a characteristic blue shift in fluorescence emission upon protein binding, as previously reported. The objective of the present experiment was production of goat embryos either by parthenogenetic activation or by in vitro fertilization and derivation of embryonic stem (ES) cell-like cells. Firstly, the effects of different activation methods, culture systems and culture media on in vitro development of zona-free and with-zona parthenogenetic embryos were investigated. Secondly, effect of cysteamine supplementation in the in vitro maturation (IVM) or culture (IVC) media, or a combination of both in terms of blastocyst production by in vitro fertilization was investigated. Finally, different methods of isolation of ES cell-like cells from in vitro produced embryos were compared and effects of Leukaemia Inhibitory Factor (LIF) and different types of feeder layers on growth of ES cell-like cells were investigated. The undifferentiated ES cell-like cells showed stem cell like morphological features, normal karyotype, expressed stem cell specific surface and intracellular markers and maintained undifferentiated state for long periods in culture. La utilización de ciertas especies arbóreas nativas andino-patagónicas con fines productivos requiere conocer sus respuestas fisiológio-morfológicas frente a factores de estrés ambiental. En el presente trabajo se estudiaron los mecanismos de respuesta de dos especies del genero Nothofagus (N. nervosa y N. obliqua). Los experimentos de estrés hídrico mostraron que estas especies poseen mecanismos anisohídricos de regulación del potencial hídrico mínimo, con un estricto cierre estomático ante altos VPD s, existiendo en N. obliqua una ventaja comparativa frente a N. nervosa al menos ante condiciones de estrés moderado. Ambas especies respondieron mejor a condiciones intermedias de sombra, siendo el principal mecanismo de aclimatación ante alta irrradiancia la producción de hojas neoformadas. En relación a la temperatura, N. obliqua mostró un mayor rango de temperaturas óptimas para la fotosíntesis, sugiriendo una mayor adaptación a condiciones climáticas variables. Los resultados de este trabajo contribuyen a entender la distribución restringida de ambas especies en el NO de la Patagonia argentina donde la variación ambiental es grande en cortas distancias. ····· 1036191247
