Fluoroquinolones are one of the new classes of antimicrobial agents that are being increasingly used against tuberculosis, the most important ones are ciprofloxacin, ofloxacin, and the new generations drugs-gatifloxacin and moxifloxacin. The clinical isolates of Mycobacterium tuberculosis (47 ofloxacin- susceptible and 71 ofloxacin-resistant) strains obtained from individual patients from various parts of India were analyzed for gyr A mutation in quinolone resistant determining region (QRDR). Most of the mutations were seen clustered in the codons 90, 94 and 95, which is a hot spot region of QRDR. The types of mutations were correlated with the in vitro susceptibility pattern of the strains to ofloxacin. The resistance to fluoroquinolones was observed predominantly due to gyr A mutations. Gyr A being a gene with house keeping functions, is highly conserved in various mycobacterial species. It was adapted for PCR-RFLP assay using gyr A gene to see the difference in the profile of 23 standard species of mycobacteria. M.tuberculosis, M.avium, and M.intracellulare produced a different profile with two different restriction enzymes. ····· 10361119609

In this study, the samples were collected from selected 40 sites of Uttarakhand (84 samples).The physico-chemical and microbial analysis was done according to standard method. The heavy metals viz. Zn, Mn, Cu, Co, Cr, As, Ni, Pb, Cd were determined in both solid and liquid wastes. The eight samples were selected, having maximum heavy metal concentrations, for the bioremediation study. The microbes were isolated from selected samples and characterized by microscopic,morphological and biochemical methods. IncP specific DNA sequences (trfA2 and oriT) were detected which revealed that IncP plasmids are prevalent in these Gram negative bacteria isolated from soil which was irrigated with wastewater discharged from industries. Present findings suggest the role of conjugative plasmids as contributors to maintenance and spread of adaptive traits (pesticide tolerance/metal and antibiotic resistance) in microbial communities. In this study, transconjugants isolated by exogenous plasmid isolation in biparental mating indicates that genes for the tolerance of pesticides and metals/antibiotic resistance can be distributed by conjugative plasmids in the natural environment. SIRT1 is a founding member of a sirtuin family of seven proteins and histone deacetylases. It is involved in cellular resistance to stress, metabolism, differentiation, aging, and tumor suppression. SIRT1-/- mice demonstrate embryonic and postnatal development defects. We examined hematopoietic and endothelial cell differentiation of SIRT1-/- mouse embryonic stem cells (ESCs) in vitro, and hematopoietic progenitors in SIRT1+/+, +/-, and -/- mice. SIRT1-/- ESCs formed fewer mature blast cell colonies. Replated SIRT1-/- BL-CFCs demonstrated defective hematopoietic potential. Endothelial cell production was unaltered, but there were defects in formation of a primitive vascular network from SIRT1-/- derived embryoid bodies. Development of primitive and definitive progenitors derived from SIRT1-/- ESCs were also delayed and/or defective. Differentiation delay/defects were associated with delayed capacity to switch off Oct4, Nanog and Fgf5 expression, decreased -H1 globin, -major globin, and Scl gene expression, and reduced activation of Erk1/2. Sanitation is the greatest medical global advance since 1840 according to a recent survey conducted by the British Medical Journal. Beaches in the Isle of Man and Cambois in the Northeast of England were designated as sites for the study of the impacts of sewage discharges from marine outfalls on the aesthetics and bacteriological quality of beaches/bathing waters. Aesthetic studies showed beaches littered with sewage derived debris, (SDD) considerably worse after heavy rainfall. The presence of SDD such as faeces, sanitary wear, floating debris, unsightly surface slicks, and discoloration are the most important features associated in the public mind with sewage pollution. Liquid and floatable solid wastes discharged at any level under seawater rise to the surface and strong winds override tidal currents in determining their direction/dispersion. Fresh sewage discharged from marine outfalls returned to pollute the study beaches/bathing waters within three hours of discharge. Culture-based enumeration methods estimated total populations of bacteria in bathing/offshore seawater samples. Total coliform and E.coli densities in bathing water samples exceeded the UK EC mandatory standards. Quantitative Rhodamine-wt tracer Dye concentrations could not be used to accurately model the dispersion of sewage bacteria in the marine environment. ····· 10361118036
