Nuclear transfer is a very effective method for propagation of valuable, extinct and endangered animals. Hand-made cloning (HMC) is an effective alternative to the conventional micromanipulator based technique in some domestic species. The present study was carried out for the establishment of an effective enucleation method, for selection of suitable somatic cell as nuclear donor and for development of an optimum culture system for in vitro culture of zona free cloned embryos of goat (Capra hircus). An efficient chemically assisted enucleation procedure was established for goat HMC. Treatment of donor cells with roscovitine resulted in better hand-made cloned blastocyst yield. Production of hand-made cloned goat embryos was better with electric pulse activation than Ca ionophore activation. A highly effective culture system for the development of hand-made cloned goat embryos was established. Furthermore, fetal fibroblast cells proved to be better nuclear donor than fresh cumulus cells or cultured cumulus cells. ····· 1036119904

The results presented here are consistent with the observation that drying and wet storage adversely affected the respiratory enzymes, PFK, MDH and NADH dehydrogenase. It is suggested that the resultant metabolic imbalance led to more leakage of electrons from the mitochondrial electron transport chain than normal, and through lipid peroxidation increased levels of hydroperoxides. However, it appears that the above biochemical events are overtaken by physical damage at higher water contents in the highly recalcitrant seeds. The present study was undertaken to determine the presence of telomerase activity in normal and cancerous cell lines and tumor tissues by combined PCR-ELISA assay. Cell lines, canine mammary tumor tissues and human oral cancer tissues were processed for protein extraction. The concentration of the protein was determined by Bradford s method and used for Telomere Repeat Amplification Protocol (TRAP). The amplified products were electrophoresed on 20% non-denaturing polyacrylamide gel along with 10bp ladder. The presence of 6 base repeats starting from 50 nucleotides is considered as positive. Then the TRAP products were immobilized on to streptavidin coated microtitre plates via biotin - streptavidin interaction and then detected by anti-DNP antibody conjugated with horse radish peroxidase. The absorbance was measured at 450 nm and at 690 nm. Out of 4 isoforms of Pyruvate kinase a glycolytic enzyme, M2 is a prototype isoform expressed in embryonic and adult dividing cells. For the first time, we reported two missense mutations in Pyruvate kinase M2 isozyme (H391Y and K422R) from the cells of Bloom syndrome patients, prone to develop cancer. Results showed that despite the presence of mutations in the inter-subunit contact domain, the K422R and H391Y mutant proteins maintained their homotetrameric structure, similar to the wild-type protein, but showed a loss of activity by 75 and 20%, respectively. Interestingly, H391Y showed a 6-fold increase in affinity for its substrate phosphoenolpyruvate and behaved like a non-allosteric protein with compromised cooperative binding. The co-expression of homotetrameric wild type and mutant PKM2 in the cellular milieu resulted in the interaction between the two at the monomer level, which was substantiated further by in vitro experiments. Interestingly, in a unique observation, hetero-oligomeric populations of PKM2 with an altered activity and affinity led to an increased rate of polyploidy i.e genomic instability. ····· 1036118638
