Although it is well established that alterations in heart rate or activation sequence induce electrical remodeling in atria, heart rate and activation sequence-induced electrical remodeling in ventricle are poorly understood. This research shows that persistent changes in ventricular repolarization can be induced by surprisingly short periods of altered rate or activation sequence, and the nature of ventricular electrical remodeling induced by these two protocols is different. This research suggests a novel mechanism for triggering electrical remodeling in ventricle. ····· 10361134187

Evidence from studies in murine and primate models show increased prevalence of Plasmodium falciparum parasitisation of reticulocytes. The increase in reticulocyte infection has been attributed to compensatory reticulocytosis and increased parasite densities. However, the prevalence of P. falciparum parasitisation of reticulocytes in human populations in holoendemic areas of malaria is unknown. Therefore, the current study examined the prevalence of P. falciparum parasitation of reticulocytes and its association with clinical, parasitological and haematological outcomes in children and adults with acute P. falciparum malaria and healthy malaria -ve controls from a holoendemic area of malaria of western Kenya. Results of this study demonstrate higher prevalence of reticulocyte parasitation in children. Thus, it seems likely that P. falciparum reticulocyte parasitation leads to a burst in compensatory reticulocytosis in acute paediatric malaria predictable by HDP and a febrile illness among children ····· 10361134089

Nutrient and waste exchanges occur through diffusion leading to low oxygen tension around the cells. Consequently even normal cartilage under normal physiological conditions suffers from a poor reparative potential that predisposes to degenerative conditions, such as osteoarthritis of the joints, with significant clinical effects. One of the key challenges in medicine is the structural and functional replacement of lost or damaged tissues. Current therapeutical approaches are to transplant cells, implant bioartificial tissues, and chemically induce regeneration at the site of the injury. None of them reproduces well the biological and biomechanical properties of hyaline cartilage. This thesis investigates the re-differentiation of chondrocytes and the repair of cartilage mediated by signaling molecules, biomaterials, and factors provided in mixed cellular cultures (co-culture systems). ····· 10361134077

Brain cancer starts in just one cell when a division starts dividing in an uncontrolled manner. As the cell makes more copies of itself, it grows to form a tumor, may be Benign or Malignant tumors. Immunohistochemical study of tissue Transglutaminase (tTG) in 22 brain tumor samples was performed. tTG expression was observed visually in endothelial and tumor cells. In most of the cases, the tTG expression was observed in endothelial cells but in few cases tTG expression was also observed in tumor cells along with endothelial cells. This shows that tumors were of high grade. We observed benign and malignant tumours like Meningioma, Papilloma, Adenoma, Astrocytoma, Glioblastoma, Multiforme Glioblastoma, Gemistocytic and Neurocytoma. Twelve females and ten males were among patients having age ranged from 8-61yrs.. The tumor were graded according to WHO grading system from grade one to grade four .Low grade tumors were more in comparison to high grade tumors. The need for a nutritionally adequate diet is of paramount importance to ensure optimum growth and development. Eating behaviours that are learned early in life are maintained in adulthood. Nutritional inadequacies influence not only adolescents` health but also risk for major chronic diseases in adulthood. As teens become more independent, they make more of their own food choices. Factors that adolescents viewed as influencing their food choices include hunger, appeal of food, time considerations of adolescents and parents, convenience of food, food availability, parental influence on eating behaviours, benefits of foods, situation-specific factors, mood, body image, habit, cost, media and vegetarian lifestyle. The present book has reviewed comprehensively key features of adolescent eating patterns which includes meal skipping, snacking, fast food consumption, inadequate intake of fruits, vegetables, and dairy and other calcium-rich foods, and unconventional dietary patterns such as adopting specific weight loss diet. ····· 10361133901

Using the hindlimb unloading model, I compared OPN-/- mice with OPN+/+ mice subjected to HU for 3 days. Whereas OPN+/+ mice suffered a marked reduction of body weight and significant spleen and thymus atrophy, OPN-/- mice were much less affected. OPN deficiency seemed to protect mice from reduction of lymphocytes in the spleen and thymus, elevation of the level of corticosteroid and apoptotic cell death. In another stress model, chronic restraint stress, I showed that OPN-/- mice are more resistant, compared to OPN+/+ mice, to CRS-induced lymphoid organ atrophy and unable to increase corticosterone production in response to CRS. ACTH also did not respond to stress in the absence of OPN. Administration of OPN into OPN-/- mice or of the anti-OPN monoclonal antibody 2C5 into OPN+/+ mice partially restored the WT and KO phenotypes respectively. Taken together, these results indicate that OPN is critical in mediating stress-induced immune organ atrophy, likely through influencing hypothalamus-pituitary-adrenal axis hormone responses. ····· 10361133708

Once cancer is formed, the prognosis is poor. The 5-year survival rate of less than 50% is one of the worst among major human cancers. One of the essential keys to improving this gloomy prognosis lies in early diagnosis and proper management of premalignant lesions. A central dogma of carcinogenesis is that alteration to critical control genes underlies malignant transformation. A major focus of this thesis was to study early molecular pathways of oral cancer development. We use Loss of Heterozygosity (LOH), a molecular analysis that can detect the phenotypic change of genome and can work on minute archival material. This thesis has not only provided a comprehensive overview of oral cancer and premalignant lesions with description on their etiological factors and clinicopathological features, but also provided data to establish molecular progression models for oral cancers, and has yielded the first evidence that premalignancies with different molecular risks could be managed differently. The results not only provide insight into the mechanism of early cancer development. ····· 10361133543

During exercise when the competition between the two metabolic pathways for the involvement of CoASH and carnitine is increased, their relative availability may play major roles in skeletal muscle fuel selection. However, the direct roles of CoASH and carnitine in muscle fuel selection during exercise have not been closely investigated in humans. This publication documents the first attempts to increase muscle CoASH content in humans, and the first demonstration that human muscle carnitine content can be increased by dietary means. Furthermore, the latter was shown to have a profound influence upon muscle fuel metabolism during exercise and exercise performance. Thus, the present data, submitted as a PhD thesis, adds valuable knowledge to our understanding of the regulation of muscle fuel metabolism during exercise in healthy humans. The findings will likely have important implications to both athletic performance as well as certain disease states where muscle metabolism is perturbed. ····· 10361133456

Red-haired people are especially vulnerable to the damaging effects of UVB due to the low content of eumelanin in their skin. Accumulated damage from UVB can lead to skin cancer. A transgenic strain of mouse containing an inducible melanogenesis system was used to investigate the mechanism of protection of eumelanin. Melanogenesis in this murine model can be induced by the application of the diterpene, forskolin to the epidermis. Forskolin increases eumelanin protection by inducing the expression of cAMP, which then upregulates the expression of melanogenic genes. To assess the protection afforded by eumelanin to the epidermis, transgenic mice were exposed to varying doses of UVB in the presence or absence of forskolin induced melanin. DNA damage was assessed by immunohistochemical staining for cyclobutane pyrimidine dimers. Vehicle treated mice and albino mice showed thymine dimer formation at 20 and 50 mJ/cm^2 of UVB. Forskolin treated and black-skinned mice showed no thymine dimer formation at these doses. These results indicate direct UVB protection by eumelanin through a light filtering process. ····· 10361132997

A biological interpretation of network alignment is to find functional homologues across different organisms. The general problem, known as the problem of finding common subgraphs across multiple networks, is NP-hard. Heuristic methods combining sequence, interaction and functional similarities have been developed to tackle it. In this book, state of the art introduction for network alignment is conducted from pairwise to multiple species. For pairwise alignment,HopMap and Mawish extension algorithms are presented with high specificity and sensitivity comparing to existing tools. The toxic effect was examined by the three major endpoints of oxidative stress. The fly mortality observed at the highest concentration of Dichlorvos indicates severity of the pesticide action on the organism. Such a drastic effect may be due the inhibitory effect of key neurotransmitter enzyme acetylcholinesterase resulting into irreparable damage to the vital cellular activities as reported earlier. The induction of key anti-oxidant enzymes Catalase and Superoxide dismutase and LPO activity indicates oxidative stress caused by the organophosphate pesticides in the exposed organism. Such a stress may arise due to generation of free radicals in the pesticide exposed flies. Another observation was that pesticide induced oxidative stress was maximum in the old flies supporting the earlier reports that aging causes more free radical generation and less quenching of free radicals.
