Drosophila melanogaster an model system for toxicity studies

The purpose of the study was to examine the currently used two organophosphate pesticides Dichlorvos and Chlorpyrifos on Drosophila melanogaster, a non-target organism. The toxic effect was examined by the three major endpoints of oxidative stress. The fly mortality observed at the highest concentration of Dichlorvos indicates severity of the pesticide action on the organism. Such a drastic effect may be due the inhibitory effect of key neurotransmitter enzyme acetylcholinesterase resulting into irreparable damage to the vital cellular activities as reported earlier. The induction of key anti-oxidant enzymes Catalase and Superoxide dismutase and LPO activity indicates oxidative stress caused by the organophosphate pesticides in the exposed organism. Such a stress may arise due to generation of free radicals in the pesticide exposed flies. Another observation was that pesticide induced oxidative stress was maximum in the old flies supporting the earlier reports that aging causes more free radical generation and less quenching of free radicals. Aldosterone is a hormone produced in the zona glomerulosa and is known to be stimulated by several hormones. ANP is known to inhibit the production of individual cells of the zona glomerulosa. This study is to see if that same affect occurs when tissue sections of the zona glomerulosa is used. Aldosterone levels were measured by an ELISA technique after 24 and 48 hours` growth, with ANP administered to the variable group after 24 hours at concentrations of 240 pg/mL and 480 pg/mL. The samples taken after 48 hours were compared to the samples taken after 24 hours, to determine a percent decrease of aldosterone production. At a concentration of 240 pg/mL of ANP, a decrease in aldosterone production of 17.4% was found in the control group versus a decrease of 25.3% in the variable group containing ANP (p = NS). However, when the concentration of ANP was increased to 480 pg/mL, the variable group decreased 54.3% compared to a 35.2% decreased in the control (p = less than 0.0005). By comparing the percent decrease of aldosterone production, a dose-responsive curve became apparent. ····· 10361132500

The purpose of this study was to conduct an investigation into the development of a soy cheese with acceptable texture. The effects of heat treatment, different oils/fat and reducing and non-reducing sugars on the soy protein isolate (SPI) and SPI curd were studied. The physico-chemical, biochemical and textural properties of three types of soy cheese were compared: (1) glucono- -lactone (GDL) induced, (2) bacterially fermented and (3) GDL-induced and fermented. The results indicate that the fermented-GDL-cheese had the most acceptable texture. Heat treatment of SPI at neutral pH caused it to denature partially and increased the storage modulus and hardness of subsequently GDL-induced gels. Subsequent heat treatment of the acidic gels further significantly enhanced the storage modulus and gel hardness. The SPI emulsion containing palm stearin showed higher emulsifying stability, emulsifying activity, protein adsorption and denaturation degree than the emulsions containing sunflower and soy oil at neutral pH. ····· 10361132299

This process may assist infection by pathogens whereas for non-pathogens apoptosis induction within carcinoma cells protects against colon cancer. Here, apoptosis induction by a major new gut bacterium, Atopobium minutum, was compared with induction by commensal (Escherichia coli K-12 strains), probiotic (Lactobacillus rhamnosus, Bifidobacterium latis) and pathogenic (E. coli: EPEC and VTEC) gut bacteria within the colon cancer cell line, Caco-2. The results show a major apoptotic effect for the pathogens, mild effects for the probiotic strains and A. minutum, but no effect for commensal E. coli. Short chain fatty acids (SCFAs) are the main fermentation products of intestinal saccharolytic bacteria in the colon and are thought to protect against colon cancer by inducing apoptosis in transformed cells. Our results confirmed that SCFAs caused morphological changes in human colonic carcinoma cell line (Caco-2) indicative of apoptosis. Analysis of various molecular markers of apoptosis (Bcl-2, caspases, cytochrome c release and cell membrane FAS levels) strongly indicated that effect. ····· 10361132056

The results indicate that Farmer Participatory Research shortens the research process and enhances acceptability and adoption of the results. However, without addressing some of the important limitations such as absence of subsidies and poor input distribution network fertiliser adoption is very unlikely.
