The type of isolates includes pathogenic group of bacteria, anaerobic organisms and other anaerobic non-spore forming Gram positive and Gram negative rods. Among the 50 samples collected, 25 contain, -haemolytic Staphylococci, 5 contain non-haemolytic Streptococci and 20 samples contain both, -haemolytic as well as non-haemolytic Staphylococci. Marine environments were investigated and 34 actinomycetes were isolated: 21 Micromonospora strains, 10 Streptomyces, and 3 Pseudonocardia. Potentially, all 21 Micromonospora strains are novel, as revealed by an original identification scheme developed to assess quickly and easily the novelty of newly isolated environmental Micromonospora strains. Standardized media for testing physiological characters of Micromonospora were developed, and additional physiological characteristics of 15 of the validly published members of Micromonospora are described. Micromonospora isolates grew under anaerobic conditions. 8 new Micromonospora species are proposed. The antibiotic biosynthetic capability of strains was assessed by PCR targeted amplification of antibiotic biosynthetic and resistance genes (oxyB, ARO-PKS, ANSA, grm, fmr). Alkaline protease producing Baciilus spp. were isolated from natural habitats of Lahore. Thirty eight isolates exhibited the prominent zones of clearance on skim milk agar medium at pH 10. These isolates which showed best proteolytic activity were then identified. Twenty two (60 %) of the alkalophilic isolates belonged to the genus Bacillus. Maximum enzyme activity (191.0U/ ml) was found in strain isolated from slaughterhouse (coded as MPPL1) at pH 10 and temperature 37°C. Maximum activity was obtained at pH 10, 140 rpm, at 37°C after 24 hours of incubation. The results indicated that protease producing acillus strain coded as MPPL1 is active at normal ranges of temperature and high pH. Alkaline protease producing conditions i.e, culture media, pH, temperature, agitation speed, incubation period were optimized at shake flask level for maximum enzyme activity and these conditions were applied at bioreactor for submerged fermentation in 1 litre jar bioreactor, an increase of nearly 80% proteolytic activity was observed after obtaining crude enzyme by centrifugation the fermented broth at 10000rpm for 10 minutes at 4°C. ····· 1036192212

A prospective cross sectional study was carried out on women who attended the family planning and gynaecologic unit of the Dschang District Hospital and other healthcare centres. This study determined the types, risk factors, pathogens of sexually transmitted diseases and other genital infections and the in vitro susceptibility profile of the most commonly isolated bacterial pathogen to ten antibiotics by a disc diffusion method on Mueller Hinton agar. Out of the 1820 women encountered in this study, 1024 ( 56.3%)women were presenting symptoms of sexually transmitted diseases and other genital infections while 796 (43,7%) were asymptomatic for these infections. Major determinants of genital infections were unprotected sexual intercourse, promiscuity, extra marital sexual practices, multiple sexuality, poverty, poor personal and environmental hygiene, frequent wearing of tight fitting clothes and synthetic underwear, the use of medicated soaps to clean the genitalia, and immunosuppression ( pregnancy and AIDS). Vancomycin (100%), gentamycin (91.7%), augmentin (87.7%), ciprofloxacin (81.3%), and streptomycin (81.0%) had high in-vitro efficacy against S. aureus strains. ····· 1036192057

Present study aimed at the detection of microbial contamination present in commercially available and freshly prepared orange juice samples collected from different localities of Lahore, Pakistan. Results showed the presence of Enterococcus spp., Streptococcus spp., Micrococcus spp., Staphylococcus spp. and Bacillus spp. in commercially available orange juices which clearly demonstrates the unhygienic conditions of processing plants. In most localities, the freshly squeezed orange juices were hygienically very poor as bacterial loads on the whole were abnormally high. Based on the presence of Enterococcus spp., Streptococcus spp., Micrococcus spp., Staphylococcus spp., Bacillus spp., Erwinia spp., Salmonella spp., Klebsiella spp., Coliforms, Pseudomonas spp., Vibrio spp. and Leuconostoc spp. in fresh orange juices, it is concluded that juices in certain areas inside the city are highly impacted and unfit for human consumption. Biodegradation of paranitrophenol (PNP) was carried out using Pseudomonas pseudomallei capable for PNP degradation. For enhanced biodegradation this strain was subjected to UV ray, chemical and gamma ray mutagenesis. UV and chemical mutagenesis was not fruitful as no PNP hyper degrading bacterial strain was obtained. Gamma ray mutagenesis was positive for PNP degradation and hyper degrading bacterial strain was obtained. Both parent and mutant in presence of glucose, efficiently degraded PNP. When concentration of glucose increased from 50 to 100 ppm then 50 ppm PNP was degraded within 12 hours by mutant and in 20 hours by parent. So glucose had positive effect on PNP degradation and resulted in enhanced PNP degradation. Effect of phenol was also checked on PNP degradation. The result showed that immobilization of the bacterial culture was best practice for PNP degradation. Similarly addition of glucose helped enhanced PNP degradation and ammonium sulphate provided good nitrogen source. After this toxicity of treated and untreated PNP was also checked by exposing to fish, that showed that after degradation of PNP no toxic metabolites were produced. ····· 1036191507

Hepatitis C virus (HCV) infection is one of the most important Flavivirida infections with significant clinical problems throughout the world in humans. Six major Hepatitis C virus (HCV) genotypes and hundreds of subtypes have been identified globally. All these genotypes are generally studied for epidemiology, their vaccine development and clinical management. This thesis comments the frequency distribution of various HCV genotypes circulate in different areas/districts of Khyber Pakhtoonkhaw Province of Pakistan. Out of the tested samples, genotype specific PCR fragments were observed in 299 (74.82%) patient serum samples. The distribution of genotypes of the typeable samples was as follows: 3 patients (0.72%) each were infected with genotype 1a and genotype 1b 240 patients (80.26%) of genotype 3a 25 patients (6.00%) genotype 3b and 28 patients (6.73%) were observed as with mixed genotypic infection. Sums of 116 serum samples (27.88%) were still found untypeable by the used molecular genotyping system. In conclusion, HCV genotypes 1a, 1b, 3a and 3b are distributed in various parts of KPK among which the genotype 3a is the most frequent genotype. ····· 1036191500

Hemorrhagic septicemia has been an acute and fatal disease among cattle and buffaloes caused by Pasteurella multocida. The prevalence of this disease is 49% in animals of Pakistan. The present study was to evaluate the prevalence, multidrug resistance and the molecular heterogeneity within clinical strains of P. multocida collected from different cities of Pakistan. Tissue samples of lung and spleen of dead animals were used for diagnostics. Rapid plate screening test for Hyaluronidase of clinical Isolates showing clear and white zones proved that given isolate is pathogenic. Amongst one hundred and ninety nine total isolates, only one hundred and one isolates were found to be virulent as clinical isolate of P. multocida. Isolates were confirmed through amplification (PCR) of specie specific and Hemorrhagic septicemia type B causing test. The primer pair of specie specific test showed a band of 300bp in all subspecies of P.multocida whereas the primer pair of HS type B causing test showed a band of 460bp in all HS-causing serotype of P.multocida. Faisalabad is known as a city of textile not only in Pakistan but also all over the world. Several textile processing units are in operation in the surroundings of the city, which are releasing a substantial amount of azo dyes into the wastewater streams in the surroundings of these textile units.The discharge of these contaminants is considered to be a potential threat to the environment because of their hazardous nature. Thus, this study was undertaken to isolate the bacteria possessing high potential to degrade azo dyes from the wastewater discharged by the textile units. A total of 150 bacterial strains were isolated from these wastewater samples using minimal salt medium spiked with Reactive Blue ZBLN azo dye. Results revealed that some bacterial strains were able to decolorize 8 different dyes almost within 6 hours. Thus, the isolated bacterial strains have the potential to remove a variety of dyes from the liquid culture, which could be used to develop a biotreatment strategy for cleanup of the industrial wastewater prior to their discharge into wastewater streams of the surrounding area. ····· 1036191291

El género Candida spp hace parte de un grupo de micosis que se pueden encontrar dentro de la biota normal de los seres humanos en sus diferentes fases dependiendo las condiciones del medio ambiente que las rodea, pero que frente a un cambio hostil en el huésped toman el papel de agentes oportunistas que se van a caracterizar por afectar a los pacientes con ciertos factores predisponentes. La aspergilosis define todos los síndromes clínicos producidos por el hongo filamentoso denominado Aspergillus, éste hongo abarca un gran espectro de enfermedades cuyo desarrollo se relaciona con las características inmunológicas del hospedero por lo cual se ha planteado que la aspergilosis es producida en su mayoría en presencia de desorden inmunológico de base. La aspergilosis afecta varios órganos, principalmente el SNC, seguido por los pulmones.
