Hospital wastewater is a reservoir of potentially harmful microorganisms. The discharge of hospital effluent directly into the municipal sewerage system or into the river without treatment could be hazardous to human health and receiving environment (water, soil and air). Randomly, triplicate hospital effluent samples at two sampling points from each 10 hospitals of Kathmandu were selected to determine bacterial load and presence of antibiotic and disinfectant resistant solates.The study showed occurrence of high bacterial load (both heterotrophic bacteria and coliform count) in all the hospital effluents under investigation, indicating high levels of pollution as safe discharge of such wastewater category should not exceed 100 CFU/ml for (TVB), 30 CFU/ml for (TC) according to the EPA Guidelines.Most of the isolates were found to be MDR.Disinfectant resistant organisms were also observed in the hospital effluent sample.Untreated liquid waste disposal into surface water directly or indirectly, has huge number effects including the addition of pathogenic microorganism, multiple drug resistance bacteria, disinfectant resistance bacteria and resistant gene in the environment. ····· 1036188244

In order to further characterise the CTL response to HTLV-1, an assay to quantify specific T cell effector functions was developed. I analysed degranulation (by CD107 mobilisation) and cytokine (IFN- and TNF- ) expression by both HTLV-1-specific CD8+ and CD4+ T cells in response to HTLV-1 Tax peptide. This study shows a trend of the frequency of degranulating CD8+ T cells to be higher in HAM/TSP patients, however at a given proviral load the rate of lysis of infected cells tend to be higher in ACs than in HAM/TSP patients. Interestingly, only the frequency of CTL degranulation correlated with the rate of killing of target cells, suggesting that CTL killing of target cells depends mainly on degranulation. ····· 1036188201

Lactobacillus species play a major role in fermented dairy products and also contribute to the therapeutic aspects of human health. Rapid methods are required for quality control of fermented dairy products, in vivo identification of probiotic strains and species of Lactobacillus and monitoring fermentation process. Though there is a fact that milk and dairy foods hampers DNA extraction due to the presence of proteins, fats and salts, Lactobacillus was identified by PCR from raw milk using modified DNA isolation technique. Incidence of lactobacillus in milk was found to be 85% among which L.delbrueckii ssp. bulgaricus was 60% and L.acidophilus was 40%. In order to validate the PCR for identification of lactobacilli from raw milk without pre-enrichment technique, the raw milk was intentionally inoculated with lactobacilli to make a concentration of 107, 105, 103, 10 cfu/ml and PCR was carried out. It concludes that at least 103 cfu/ml of lactobacilli is required to identify the lactobacillus from raw milk by PCR. ····· 1036188135

Two DC methods AO and DAPI, were optimised and applied to quantify total populations of bacterial cells in natural waters, with expected differing bacterial numbers, nutritional contents, temperature and salinity. Various culture-based methods were also applied to quantify viable bacterial populations. The two DVC methods optimised in this study, successfully quantified `Living` (Viable culturable and VBNC) bacterial populations in natural waters. NADVCs and INTDVCs were consistently lower than total direct counts and also consistently higher than standard heterotrophic plate counts. Results obtained for cultured bacteria using both DVC methods were similar. However, for mixed bacterial populations, the counts were always higher with the INT method than with the NA method. Combining both DVC methods yielded a statistically significant increase in numbers of INT active cells compared with the INT method used alone. Substrate-enhanced INT method when combined with digitised image analysis, allowed for the identification of different bacterial morphological types and the quantification of the overall physiological states of individual bacterial cells in samples of raw domestic sewage. ····· 1036188000
