DNA Profiling of Chilli (Capsicum annuum L.)

Analysis of genetic diversity is useful in selecting diverse parental combinations, reliable classification of accessions,for exact identification of variety, for conservation and utilization of plant genetic resources. In the present work genetic diversity in 23 chilli genotypes was analyzed by RAPD markers generated by 13 random primers out of which 9 were polymorphic. The amplification profile of these 9 primers consisted of 63 fragments of size ranging from 391 bp to 2754 bp of which 35 were monomorphic and 28 were polymorphic with 44.4 % polymorphism. The number of bands generated by each primer varied from 5 (OPB-10) to 9 (OPB-01 and OPB-15) with an average of 7 fragments per primer. The percentage of polymorphic bands with different primers ranged from 14.29 to 66.67 %. The similarity coefficients detected by RAPD ranged from 0.42 to 0.97 which revealed existence of limited genetic variation among chili genotypes. The consensus tree constructed showed three major clusters. First cluster comprise of 13 genotypes, second cluster of 7 genotypes and third cluster of 3 genotypes. The RAPD technology proved useful in describing genetic diversity among chilli genotypes. ····· 1036192611

Septoria leaf spot caused by Septoria lycopersici Speg. and late blight caused by Phytopthora infestans (Mont.) de Bary are two important foliar diseases of tomato (Solanum lycopersicum L.). Identification of DNA markers linked to these diseases is a prerequisite for accelerating resistance breeding through marker-assisted selection (MAS). The objectives of the present study were to investigate the inheritance pattern of septoria leaf spot resistance and identify molecular markers linked to septoria leaf spot and late blight resistance in tomato. The inheritance of resistance to septoria leaf spot was based on a single dominant gene. Transgressive segregation for resistance and susceptibility to late blight was also observed. Forest Cover Change Detection Using Remote Sensing and GIS Techniques

Forest cover change analysis between 1973 and 2003 was conducted in Banja Woreda, Awi zone, Amhara region using remote sensing and geographic information system (GIS) supported with field verifications. Information was extracted from various Satellite images and different digital maps. The objectives of this study were to detect the extent and rate of forest cover change over the last 30 years. Three dates of Landsat image data of the 1973, 1986 and 2003 were used to produce land cover map in general and forest cover map in particular. NDVI image differencing and post-classification comparison change detection methods were employed. In addition to this, household levels of socioeconomic data were used in explaining the causes of forest cover changes in the study area. The results show that during the last 30 years, forest cover declined from 6044 ha in 1973 to 2855.9 ha in 1986 and 2446.9 ha in the year 2003. The annual rate of forest cover change between 1973 and 2003 was 120 ha/year. The socio-economic factors like population growth, the demand for the expansion of agricultural land, fuel wood and construction materials were major driving forces for observed forest cover changes. ····· 1036192230
