Prebiotic galacto-oligosaccharides (GOS), which are non-digestible carbohydrates and are produced by transgalactosylation of lactose catalyzed by galactosidase, have been found to modulate the colonic micro flora by stimulating the growth of probiotics and suppressing adverse microbial activities and have received much attention in last few years. The prebiotic efficacy of GOS is largely dependent on glycosidic linkages among monosaccharides and GOS mixtures typically result in better growth of that strain that had served as galactosidase source. In the present book, galactosidases from different lactobacilli and other sources have been characterized in detail for various biochemical properties and their transgalactosylation abilities have been evaluated. To overcome the problem of less activity, genes of galactosidases were cloned and expressed in different expression systems. The experiment aimed to study the effect of drip fertigation at different levels of fertilizer dosage in both small onion (COOn5) and big onion (Agri Found Dark Red). This study concerned about the usage of low cost straight fertilizers. If there is a possibility of using easily available water soluble straight fertilizer through drip, then a drastic cut in the cost of cultivation can be achieved. Total Bulb yield and Quality were increased with the increase of N and K levels but only upto a certain level. When the fertilizer dosage exceeds high (125% of recommended dose of fertilizers) there is a reduction in yield. The study revealed that application of 75 per cent RDF (i.e., 75:112.5:56.25 kg NPK ha-1 in big onion and 45:45:22.5 kg NPK ha-1 in small onion) as fertigation proved its superiority over other treatments in respect of growth, yield and quality traits. ····· 1036149

A significant part of sugarcane crop is lost each year due to different diseases particularly red rot. As sugarcane is mostly propagated by vegetative method, therefore, plant once infected by pathogen can transfer it generation after generation. Therefore introduction of new disease resistance varieties needs rapid, reliable and efficient methods of multiplication. In this study the, biotechnological methods i.e. Plant Tissue Culture were used for genetic improvement through both physical and chemical mutagenic agents. In this project the major emphasis was focused on in vitro as well field screening of red rot resistant lines using partially purified toxin of fungus Colletotrichum falcatum. The induction of mutation was further supported by using various disease related biochemical markers(Qualitative and quantitative analysis of protein and different enzymes). Finally these selected high yielding red rot resistant lines were multiplied by micropropagation, regeneration through callus to achieve the objectives. Protocols are suitable for induction of mutation and mass scale production of disease resistance, high yielding and premium quality planting material. ····· 1036147

Pakistan has the largest contiguous gravity flow canal irrigation system, but is falling short of good-quality water due to increased cropping intensity and increased demands for households and industry over the years. In Pakistan the agriculturally important areas are arid or semi-arid where freshwater supplies are limited to meet crop water requirements especially at critical stages. This shortage is being fulfilled by exploiting groundwater resources which is mostly brackish in nature. We evaluated the growth response of wheat and berseem crops to brackish water during reclamation of dense saline-sodic soils. The cyclic irrigation of CW and TW along with the application of gypsum @ 100% SGR or FM or with auger hole 60 m-2 refilled with gypsum and rice husk (1:1 ratio) remained better in lowering ECe, pHs and SAR and produced maximum grain and straw yields of wheat as well as berseem fodder. Overall, it was concluded that low quality waters could be exploited for irrigation on marginal salt-affected soils. ····· 1036135

The crop water requirements for the major Rabi crops viz. wheat, barley and gram were computed by Penman-Monteith method using computer program `CROPWAT`. Distributary wise water supply and water demand was estimated for water balance analysis. The performance was evaluated using the adequacy and relative water supply indicators. These indicators were calculated for the ten distributaries for the 5-month irrigation period of Rabi season. Data ware collected from field and irrigation department. The analysis of results indicated inadequate and inequitable canal supply mainly at tail distributaries of both LMC and RMC. A linear programming irrigation planning model was developed considering three specific objectives viz. production maximization, net benefit maximization and labour employment maximization and is experimented for various levels of surface water availabilities i.e. no of canal running days in a season (70,80,90,100,110 & 120days).
