This thesis focuses on how the cholesterol content of the cell membrane effects the temperature activation of the TRPV1 ion channel. TRPV1 is a temperature activated ion channel, which acts as a heat pain sensor in peripheral nervous system and also plays an important role in inflammatory pain sensing (heat hyperalgesia). Results showed that untreated cells were activated at about 42°C which confirmed the results from other studies, whereas the activation temperatures of cholesterol depleted and enriched cells were both significantly increased. For the last 25 years, El-Asmer and co-workers were pursuing the search for potential therapeutic agent or new drug models from snake venoms. This work was presented for partial fulfillment of M.D. in Biochemistry. A cytolytic, non-antigenic, 8 kDa, glycoprotein (Fraction VIb) was isolated from crude Naja nigricollis nigricollis spitting cobra venom. This protein is stable at different pH values and different temperatures up to 100ºC, lethal to mice, with LD50 1.83 mg/kg. Small doses induced cytolytic effect on hepatocellular carcinoma, colon cancer and breast cancer cells while larger doses caused hemolysis of red blood cells. Lectins are carbohydrate-binding proteins of non-immune origin that agglutinate cells or precipitate glycoconjugates.Based on evolutionary and structural relatedness, seven lectin families have been distinguished, of which legume lectins are the best characterized. Lectins typically constitute a large fraction of the total protein in most leguminous seeds and plant storage tissues where they play an important role in several defense mechanisms. A lectin from the seeds of Indigofera heterantha, member of family leguminosae was isolated, purified, partially characterized and its antimicrobial activity evaluated. The lectin is a homotetramer with a molecular weight of about 70 KDa. The lectin shows a reasonable temperature stability and is stable in the pH range of 2-9. It demonstrates a remarkable antibacterial activity against E.coli, K.pneumonia, S.aureus, and B.subtilis and also inhibits the growth of phytopathogens Aspergillis niger, Aspergillis oryzie and Fusarium oxysporum. Though considerable progress has been made on the contraception among females, progress on males are still limited. Effects of different treatments on various male reproductive endpoints and contraceptive efficacy were evaluated. Histopathology revealed that marked atrophy of the seminiferous tubules characterized by disruption of the germinal epithelium and atrophy of the leydig cells as well as epididymal sperm counts were reduced drastically. Hormonal assay revealed a significant decrease in serum level of testosterone, FSH and LH resulting in suppressed spermatogenesis of male rats. These effects lead to the reduction of pregnancy rate of untreated fertile female rats while mated with the extract-treated male rats. The results showed that A. precatorius seed extract causes suppression of spermatogenesis and fertility, thereby suggesting the potential of this plant in the regulation of male fertility. ····· 1036188400
