Gram-negative bacteria have evolved an array of secretion systems for protein transport across their membranes. The ability to secrete a diverse set of proteins to their target of choice provides bacteria with a menacing tool for pathogenesis. This work aims to enhance our understanding of bacterial pathogenesis by characterizing key aspects of bacterial secretion systems using mass spectrometry. Chapter 1 provides a general introduction of protein secretion pathways in Gran-negative bacteria, with a special emphasis on type III and type VI secretion systems. Chapter 2 highlights chemical cross-linking and mass spectrometry (CXMS) as a technique for studying protein-protein interactions in large molecular complexes. Chapter 3 describes a novel pipeline for acquisition and analysis of CXMS data. In Chapter 4, key interactions between the basal components of type III secretion systems (T3SS) are identified, and used to create a model of the core structure of T3SS. In Chapter 5, secretome of T6SS in P. aeruginosa is profiled to identify three novel substrates of the system. Proteinase inhibitors (PIs) are small proteins that are quite common in nature. PIs are present in multiple forms in numerous tissues of animals and plants as well as in microorganisms. Serine proteinase inhibitors are widespread in the plant kingdom. The present study was at investigating the biochemical properties of protease Lavatera cashmeriane seeds. Four protease inhibitors LC-pi I, LC-pi II, LC-pi III and LC-pi IV were purified from the seeds of Lavatera cashmeriane by ammonium sulphate precipitation and ion-exchange chromatography on DEAE-cellulose column. All were strong inhibitors of trypsin, chymotrypsin and elastase. The molecular weight of LC-pi I, LC-pi II, LC-pi III, and LC-pi IV was found to be 20.89, 14.12, 16.78 and 7.94kDa respectively by gel filtration chromatography and 10, 14, 16 and 7kDa respectively by SDS-PAGE. The SDS-PAGE revealed that LC-pi I is constituted of two subunits of 10,000 Da each. The optimum temperature for the inhibitors was found to be 30 C for all inhibitors. LC-pi I showed strong antibacterial activity against Klebsiella pnuemoniea, and Pseudomonas aeruginosa but was less active against E.coli. ····· 1036199583

The glyoxalase system is made up of two enzymes that detoxify methylglyoxal and the other reactive oxoaldehydes, which are produced as a normal part of metabolism. In this book, the characterization of two GLX2-like isozymes from Arabidopsis thaliana and human GLX2 is presented. Our work demonstrates that GLX2-1 is not a glyoxalase 2 however, the protein does contain a dinuclear metal center. We found that GLX2-1 does catalyze the hydrolysis of beta-lactam antibiotics. Overall results suggest that different isozymes of GLX2 have different metal centers and different activities. Also, both the highly conserved metal binding domain (T-H-X-H-X-D-H) and the substrate binding ligands need to be concerned to be considered when classifying glyoxalase 2 enzymes. The results in this book further our understanding of the GLX2 enzymes. It has helped elucidate the structure, function, and mechanism of these enzymes. Our results are contributing to the ultimate goal in glyoxalase research in search of inhibitors that can be used as potential drugs in the treatment of glyoxalase system-related diseases, and the results also suggest that this enzyme may be the template for evolving new functions ····· 1036199557
