Rising human population throughout the world has detrimental effects on life supporting system on earth. Though considerable progress has been made on the contraception among females, progress on males are still limited. Numerous plants have been used historically to reduce fertility in male and modern scientific research has confirmed contraceptive potentials of Abrus precatorius seed extracts. Effects of different treatments on various male reproductive endpoints and contraceptive efficacy were evaluated. Histopathology revealed that marked atrophy of the seminiferous tubules characterized by disruption of the germinal epithelium and atrophy of the leydig cells as well as epididymal sperm counts were reduced drastically. Hormonal assay revealed a significant decrease in serum level of testosterone, FSH and LH resulting in suppressed spermatogenesis of male rats. These effects lead to the reduction of pregnancy rate of untreated fertile female rats while mated with the extract-treated male rats. The results showed that A. precatorius seed extract causes suppression of spermatogenesis and fertility, thereby suggesting the potential of this plant in the regulation of male fertility. ····· 1036188400

Studies described in this dissertation have focused on understanding the crosstalk between two different classes of membrane proteins: Receptor and non-receptor tyrosine kinases and gap junctions. The first part of the study was focused on the non receptor tyrosine kinase pp60v-Src, and the various signaling pathways it utilizes to acutely inhibit Cx43 gap junctional communication. Here we have identified the involvement of at least three signaling pathways which co-coordinately regulate the acute inhibition of Cx43 coupling. The second study was directed towrards understanding the underlying principle for the transient shut down of Cx43 gap junctional communication by epidermal growth factor. Here we generated a stable clonal pool of HeLa cells expressing a growth factor insensitive mutant of Cx43 that was lacking targets for ERK phosphorylation. This study aimed to evaluate the chemopreventive and antioxidative effects of Eleusine indica and Thysanolaena latifolia against CCl4-induced liver damage. Rats were pretreated with E. indica and T. latifolia for fourteen consecutive days prior to the administration of CCl4 (two doses). Serum levels of alanine aminotransferase (ALT), aspartate aminotransferase (AST), malondialdehyde (MDA), glutathione (GSH), antioxidant enzymes and histopathological changes in rat liver were determined. Total phenolic content of E. indica and T. latifolia were found to be 14.9 ± 0.002 and 20.3 ± 0.007 mg/g total phenolic expressed as gallic acid equivalent per gram of extract respectively. The extracts were able to reduce the stable DPPH in a dose dependent manner. The IC50 value for E. indica and T. latifolia were found to be 2350 µg/ml and 1525 µg/ml. However, most of these changes were alleviated by prophylactic treatment of animals with E. indica and T. latifolia dose dependently. We concluded that E. indica and T. latifolia could be used as hepatoprotective agents and possess the potential to be used to treat or prevent degenerative diseases where oxidative stress is implicated. ····· 1036187568

First cadmium metallothionein like gene (PMCd1) from a ciliate, Paramecium sp. and a novel cadmium inducible metallothionein (TMCd1) gene has been identified and sequenced from the locally isolated ciliate Tetrahymena tropicalis lahorensis isolated from industrial wastewater. Both the genes (PMCd1 and TMCd1) are intronless, encoding 612 and 471 nucleotides respectively, with TGA as the stop codon and TAA coding for glutamine. The coding region of PMCd1 comprises 203 amino acids, including 37 cysteine residues with a conserved structural pattern arranged in 17 cys-x-y-cys, 1 cys-cys and cys contexts. The TMCd1 is quite different from the previously reported MT genes in T. pyriformis and T. pigmentosa. However, it shows 78% homology with four different Cd-MT genes reported from T. thermophila. The amino acids sequence of TMCd1 has a special feature of three CCCX6CCX6CCCX6CC and two CCX6CXCX2CXCC intragenic tandem repeats with a conserved structural pattern of cysteine. The translated protein of TMCd1 contains 30.12% cysteine residues, which is typical of Tetrahymena Cd inducible MT genes. Also there was significant decrease in serum levels of triglycerides (TG), total cholesterol (TC) , low density lipoprotein cholesterol (LDL-C) and a concomitant increase in high density lipoprotein cholesterol(HDL-C) in Aloe vera, Fenu Greek and Glibenclamide treated groups. Fenugreek treated groups showed a significant decrease in TC, TG, LDL, and VLDL when compared to Aloe vera treated group. Das Teilen von Nahrung ist kein seltenes Phänomen unter Primaten. Es wurde unter anderem bei Kleideraffen, Gibbons, Schimpansen und auch bei den in dieser Studie behandelten Bonobos beobachtet. In den zuerst genannten Fällen kann dieses Verhalten meist auf Basis der Verwandtschaftsselektion erklärt werden. Betrachtet man jedoch das Teilen von Nahrung bei Bonobos, treten zwei Besonderheiten in den Vordergrund. Erstens teilen am häufigsten die aufgrund der patrilinearen Gruppenstruktur nicht oder nur entfernt verwandten Weibchen die Nahrung miteinander. Dies lässt die Frage aufkommen, welche Faktoren dieses Verhalten zwischen nicht-verwandten Tieren begünstigen. Zweitens tritt das Teilen von Nahrung oft zeitnah zu sexueller Interaktion auf. Hierzu beschreibt z.B. Frans de Waal (1998), einer der renommiertesten Forscher im Bereich Verhaltensforschung bei Primaten, Situationen, in denen die Bereitschaft eines Männchens zum Teilen von Nahrung mit einem Weibchen durch eine vorangegangene Kopulation positiv beeinflusst zu sein schien. Sowohl Weibchen als auch Männchen forderten dabei zur Kopulation auf, wobei Weibchen anschließend stets von der Nahrung des Männchens fressen konnten. Seine Beobachtungen interpretierte de Waal (1998) dahin gehend, dass Männchen Nahrung gezielt dazu einsetzen, um mit Weibchen zu kopulieren und Weibchen ihrerseits die sexuelle Attraktion der Männchen nutzen, um Zugang zu einer begehrten Nahrung zu erlangen. Nun kann dieses Verhalten in heterosexuellen Paarkonstellation leicht durch die Begrenzungsfaktoren des reproduktiven Erfolges beider Geschlechter erklärt werden, welche für Weibchen durch den Zugang zu hochwertiger Nahrung und für Männchen durch den Zugang zu fruchtbaren Weibchen bestimmt sind. Amy Randall Parish (1994) beobachtete jedoch ein ähnliches Verhalten zwischen Weibchen untereinander. Wenn Weibchen die Nahrung miteinander teilten, kam es dabei ebenfalls häufig zur sexuellen Interaktion in Form des für Bonobo-Weibchen typischen Genito-Genital-Reibens (GG-rubbing). Auch Parish vermutete einen engen Zusammenhang zwischen dem Teilen von Nahrung und dem Auftreten von Genital-Kontakt entsprechend eines Austausches von sexueller Interaktion und Nahrung zwischen den beteiligten Tieren (`Sex-for-Food-Exchange`). Sie konnte in einer experimentellen Studie an in Gefangenschaft lebenden Bonobos zeigen, dass dieser `Sex-for-Food-Exchange` regelmäßig unter Weibchen vorkommt. Bisher wurden jedoch keine Daten über den zeitlich direkten Zusammenhang von sexueller Interaktion und Nahrungsteilen zwischen weiblichen Bonobos erhoben. Ziel dieser Studie ist es deshalb, weitere Erkenntnisse über die Funktion von GG-rubbing im Sinne eines direkten `Sex-for-Food-Exchange` zwischen Bonobo-Weibchen zu erlangen. ····· 1036173874

Endometrial carcinoma is one of the most common malignancies in the female genital tract and poses serious health hazards. The gene P53 safeguards cell integrity however a mutation in this gene increases the lifeline of the cells which are programmed for cell death. This particular gene participates in many human cancers including endometrial, ovarian and tubal carcinomas. In this study we screened exon 5 of p53 gene to find out whether this exon is a hotspot for mutations or not. We evaluated (n=45) samples of endometrial carcinoma and (n=45) controls from a local population. However, we were unable to find any nucleotide alteration in exon 5. It appears, therefore, exon 5 of p53 is not a hot spot for mutation in local population based on recent our findings. ····· 1036125953
