Anti-inflammatory and antimicrobial properties of Ficus exasperata

The present work describes the investigation of extracts of the leaves and stem bark of F. exasperata for anti-inflammatory, anti-oxidant and antimicrobial activities as suggested by folklore medicine. It also describes the isolation and characterisation of the bioactive principles from F. exasperata. Anti-inflammatory activities of the extracts and isolates were investigated using the carrageenan-induced foot pad oedema model in the seven-day old chicks. Antioxidant protocols used included the DPPH scavenging,linoleic acid lipid peroxidation and ferric reducing power assays. The agar well diffusion and micro-dilution assays were used in the antimicrobial studies. The active principles were isolated by using column and thin layer chromatographic techniques and characterised by their NMR spectra data. The stem bark extract showed considerable anti-inflammatory, antioxidant and antimicrobial activities. Bergapten and oxypeucedanin hydrate, isolated from the stem bark, sitosterol and sitosterol-3-O-beta-D-glucopyranoside, isolated from the leaves contributed to the bioactivities of F. exasperata. Motor incoordination was evaluated using inclined-beam walking test.Pretreatment with onion extracts reduced cerebral infarct size, attenuated impairment in short term memory and motor coordination along with a decrease in mitochondrial TBARS. Die Sprühgefriertrocknung (SFD) ist ein modernes Verfahren um proteinhaltige Mikropartikel herzustellen. Unter SFD versteht man eine Kombination aus der gewöhnlichen Gefriertrocknung und einem vorgeschaltetem Vernebelungsschritt der Proteinlösung in flüssigem Stickstoff. Durch niedrige Temperaturen und Unterdruck im Gefriertrockner wird das Eis der gefrorenen Pulverpartikel durch Sublimation getrocknet und es entsteht ein feines Pulver sphärischer Partikeln mit definierter Partikelgröße. Die Qualität und Unversehrtheit solcher Proteinzubereitungen kann stark durch unterschiedliche Faktoren während der Herstellung beeinträchtigt werden. Der Schwerpunkt dieser Arbeit war die Untersuchung von prozessbedingten Schädigungen der Proteinstruktur mit Hilfe eines Mehrwinkel-Lichtstreudetektors (MALS) und die Steuerung der Partikelmorphologie von SFD-Pulvern hoher Dichte. Das Arbeiten und die Vorzüge des eingesetzten Drei-Detektoren-Systems bestehend aus UV-DAD, MALS und RI-Detektor wurde näher erläutert (Kalibrierung, Normalisierung, Berechnung der absoluten Masse der zu untersuchenden Proben). Es konnte gezeigt werden, dass Molekulargewichte von Proteinen durch LS genauer errechnet werden können als es mit einer Kalibriergerade der Fall ist, zumal dieses Verfahren unabhängig von der Flussrate sowie der Zusammensetzung der (isokratischen) mobilen Phase ist. Bei unveränderten Versuchsbedingungen wurden Brechungsindexinkremente, Absorptionskoeffizienten sowie sekundäre Virialkoeffizienten bestimmt. Neben der SEC (Size exclusion chromatography) wurde die AF4 (Asymmetrical flow field-flow fractionation) als weiteres analytisches Verfahren zur Proteincharakterisierung etabliert. Bovine Carboanhydrase (bCA) wurde als Modelproteine für die SFD verwendet. Versuche mit 15 %igen bCA Lösungen lieferten ein umfassendes Bild über die Proteinschädigung der einzelnen Prozessschritte. Während die Zerstäubung der Lösung nur geringfügigen Schaden hervorrief, kam es beim Lösen hoher Konzentrationen und v.a. der Trocknung zu beträchtlichen Aktivitätsverlusten. Auffallend war eine Rückgewinnung der Enzymaktivität nach der Vernebelung von bCA in flüssigem Stickstoff, welche in der Literatur auch für andere Proteine beschrieben ist. Versuche mit unterschiedlichen Feststoffgehalten haben gezeigt, dass mindestens 5 %ige Feststofflösungen nötig sind, um runde Pulverpartikel herzustellen. Durch Variationen der Prozessparameter konnte ein umfassendes Bild zur Enzymschädigungen während der SFD gewonnen werden. Durch den Einsatz unterschiedlicher Zusatzstoffe konnte das Modellprotein vollständig stabilisiert werden. Auch bei der Sprühtrocknung (SD) von bCA waren hohe Aktivitätseinbußen zu beobachten, welche vergleichbar mit den Ergebnissen der SFD waren. Pulver die für die nadelfreie Injektion in die Haut (EPI = epidermal powder injection) eingesetzt werden müssen eine bestimmte Partikelform / -größe, sowie eine hohe Dichte aufweisen. Durch den gezielten Einsatz von Polysacchariden während der SFD von bCA und Albumin (bSA) konnte eine Schrumpfung und somit Verdichtung der Pulverpartikel herbeigeführt werden. Annealingversuche mit hilfsstofffreien Formulierungen zeigten keine sichtbare Veränderung der gefriergetrockneten Pulverpartikel. Temperaturerhöhung und verlängerte Annealingdauer führten zu keiner Verdichtung der Pulverteilchen. Erst eine Temperaturüberschreitung von Tg¿ während der Primärtrocknung erzeugte kollabierte Partikel. Quecksilberporosimetrie und BET-Messungen bestätigten die Ergebnisse der REM-Aufnahmen. Im Vergleich zu vorhergehenden Versuchen mit kurzen Gefriertrocknungsprogrammen hatten die geschrumpften Pulverpartikel weniger/keine Poren und nur etwa ein Drittel der ursprünglichen Gesamtoberfläche. Aktivitätsmessungen von redispergierten, geschrumpften bCA Partikel zeigten, dass die Proteinstruktur von kollabierten Pulverpartikeln zusätzlich geschädigt wurde. ····· 10361107275

There are 5 major periods of concern when considering use of an antimicrobial agent, or other drug, during pregnancy. The preconception period is important for both males and females because drug administration to either gender in animal models can affect embryonic development or result in embryonic death. Use of drugs during the first 7 days after conception probably represents an all-or-none effect on the fertilized ovum: Either the damage is so great that the embryo dies, or the damaged cells are replaced by undifferentiated cells that develop normally. Once implantation occurs, after 14 days, differentiation of the embryo begins. This is the period of time when most drug-associated congenital abnormalities are produced.
