It was found that the cumulative % release of drug decreased with the increase of polymer load in all cases. Better sustaining effect was found from Eudragit RS 30 D and xanthan gum combinations. In pylorus legated rats, a significant reduction in free acidity, total acidity and ulcer index was observed with high dose while low dose produced reduction in free acidity and ulcer index. Both doses also showed significant antiulcer effect in ethanol-induced and stress-induced gastric ulcers. Losartan also reduced ulcer area in cysteamine-induced duodenal ulcer. The results indicated that gastric mucosal damage was significantly reduced by clonazepam, as well as by Ro 15-4513 (partial BZR inverse agonists) pretreatment, significantly reversed by flumazenil (a central BZR antagonist). CGS 9896 (a partial agonist at the BZRs) and Ro 5-3663 (an atypical BZ, which bind to the picrotoxin site) did not offer any protection against ethanol.
