The present study is to investigate the effect of Ammonio Methacrylate Copolymer Type B (Eudragit RS 30 D) on the release kinetics study of diclofenac sodium form coated pellets. Eudragit RS 30 D and xanthan gum was added into the formulation at 5:1 ratio. Different percent of this polymeric combination was loaded on to the drug-loaded pellets. It was found that the cumulative % release of drug decreased with the increase of polymer load in all cases. Better sustaining effect was found from Eudragit RS 30 D and xanthan gum combinations. The effect of centrally administered losartan, an angiotensin-2 receptor antagonist on gastric acid secretion and gastric cytoprotection was studied using different models of gastric ulcers such as acetic acid-induced chronic gastric ulcers, pylorus ligation, ethanol induced and stress induce acute gastric ulcer and cysteamine hydrochloride induced duodenal ulcer. Losartan was administered intracerebroventrically at two different doses of 125 micro gm/kg and 250 micro gm/kg. Both doses of losartan increased healing of ulcer in acetic acid induced chronic gastric ulcer model. In pylorus legated rats, a significant reduction in free acidity, total acidity and ulcer index was observed with high dose while low dose produced reduction in free acidity and ulcer index. Both doses also showed significant antiulcer effect in ethanol-induced and stress-induced gastric ulcers. Losartan also reduced ulcer area in cysteamine-induced duodenal ulcer. It was concluded that angiotensin II receptor antagonism in brain increases healing of gastric ulcers and also prevents development of gastric and duodenal ulcers in rats. ····· 10361147028

Benzodiazepines (BZs) were reported to protect against ethanol-induced gastric damage in rat s in vivo and in vitro model. However, the effects of some drugs in the new in vitro model do not agree with the results reported before in vivo studies. Therefore, the present study was designed to modify the new in vitro model and to assess the gastroprotective effects of some BZs, and also some new compounds. Gastric mucosal damage was induced by instillation of ethanol into the isolated rat stomach kept in aerated Kreb s solution at 37°C in an organ bath. Drugs or their vehicles were administered inside the bath 15 min before ethanol. The results indicated that gastric mucosal damage was significantly reduced by clonazepam, as well as by Ro 15-4513 (partial BZR inverse agonists) pretreatment, significantly reversed by flumazenil (a central BZR antagonist). CGS 9896 (a partial agonist at the BZRs) and Ro 5-3663 (an atypical BZ, which bind to the picrotoxin site) did not offer any protection against ethanol.
